I would suggest searching this forum for ideas, there are literally thousands of postings with hundreds of ideas on what to see. I would suggest obtaining a good guidebook for Berlin (I like DK Eyewitness Berlin), read it cover to cover, come up with some things you want to do, then look to the experts on this forum for more information. Berlin has a lot to offer, it depends on your interests. Search the forum for info on bike and Segway tours, it's here.
On your first day of arrival you could avail yourselves of SANDEMAN free tours which operate on tips basis. Thus, you will be able to get some general idea of the city and main historic and cultural sites. And then you could ask a guide for some ideas what to see, what to do or where to go.

Please use another tour company besides the Sandemann tours. This theme comes up on here on this forum all the time. There are many other tour companies that are far more reputable than the "New Europe" free tours.
Read the reviews on the Things to Do in Berlin page for the best tours. Go to their websites to see what tour best matches your schedule and interests.
For a good tour-like trip around the city, you could hop on Bus 100. It loops around the major sight in Berlin. Some top things to see in Berlin, that I would recommend for a first time visitor would be the Reichstag (German Parliament building) and at least one of the musuems on Museum Insel (my personal favorite Pergamon museum). Some nice areas of the city include Unter den Linden, the Gendarmenmarkt and the KuDamm. Also, the palaces in Potsdam (Sansoucci and the Neue Palais) are an easy day trip via public transporation from Berlin, if you'd like to get out of the city for a bit.
If I got a bus 100 would it eventually get back to where I got on or would I have to change at say a terminus?
If I hopped off somewhere that took my fancy could I just keep jumping on a 100 bus.What I mean is does it keep doing a circular route?
Bus 100 route is served in both directions in 10 minutes intervals.
But you can make a round trip by using the bus 200. Like the 100 bus line the 200 does also connect many tourist sights. Thus you can make a trip: Berlin Zoo station - bus 100 - Berlin Alexanderplatzstation - bus 200 - Berlin Zoo station.

As you've got 4 days: another good bus route is the M29. Check out this article:
S-Bahn line between Zoo and Alexanderplatz is also great for views. Boat trip in central Berlin is also good esp. to see new government buildings.
