We stayed at the Squires Guest House in early March, yes the weekend that the whole of the UK was buffetted by a huge storm including gale force winds that came directly through Plymouth.
The Guest House itself is nice, in a quiet location that's fairly central and only a very short walk from Plymouth Ho, and about 5 minutes or so from the area with bars, restaurants etc.
Driving directions on their website are good, despite the confusing one way system in the middle of Plymouth! Parking is a challenge due to city-centre pay-and-display bays being everywhere but for a very modest fee you can get all day parking and it's free between 18:00 and 10:00 the next day.
The other reviews mentioned a great breakfast at this hotel and we can certainly vouch for that! Very high quality and hand-cooked to order. Also the owners are very friendly and go out of their way to be helpful, making it an absolute pleasure to stay there.
