Buses only go to the entrance to the city and not around city streets. To get to a destination or hotel within the city you can use a taxi (or horse-drawn vehicle by day) but be aware that there are lots of slopes up and down in the city area and whilst walking down the main street to the main square is fairly gentle, roads off are quite steep.
Hope you don't miss the chance to visit Valletta - as a Brit I like it immensely..
