upgrading cabin class after ticket has been bought with BA

In order to be able to upgrade cabin class on an existing booking with BA, using either MFU or by paying, must you have booked the orginal ticket with "flexible conditions" or can you still upgrade if you have purchased a "lowest available" ticket?

Depends on the cabin class.

If you bought World Traveller or Euro Traveller tickets then it must be a semi-flexible ticket to be able to upgrade. Any paid ticket in World Traveller Plus or Club World is upgradeable with miles. HOWEVER THIS IS ONLY IF YOU BOUGHT IT FROM BA. Sorry for the capitalisation but that is important!
