ESTA question

We are flying from Glasgow (Scotland) to Las Vegas, via Philadelphia, where we change planes.

I am about to update my Esta with our trip details, but I'm not sure if I should put down the Phili flight number or the Vegas flight number.

I presume we will be clearing customs/immigration in Phili. Flying with US Airways.

Yep - on entry to the US, you clear both immigration and customs at your first point of entry, in this case PHL - so that's the flight number to put down (although that section is optional in any case).
