You should then see an evening departure with 1 change marked CNL, ICE. This is the Venice to Munich City Night Line sleeper train, then an onward high-speed ICE train next day.
In principle, fares start at 59 euros with a couchette or around 90 euros with a bed in a 3-bed sleeper, but fares vary depending on date and how far ahead you book. Booking usually opens 90 days ahead.
For Istanbul, book Frankfurt to Vienna on the late night sleeper from 39 euros with couchette or 79 euros with bed in 2-bed sleeper. Then book Vienna to Budapest from 19 euros also at www.oebb.at. Then book the sleeper from Budapest to Bucharest at www.mav-start.hu from around 49 euros with bed in a sleeper.
Finally, buy a Bucharest to Istanbul ticket on the daily 'Bosfor' train at the station when you get to Bucharest, fare around 40 euros plus 9 euros for a couchette or 20-odd euros for a bed in a 3-bed sleeper.
I am a bit scared to do so many changes for train travel from frankfurt to istanbul. Is it a long trip?
Yes, it is a very long trip. Expect it to take over three days that way, and you will need to be a real train fan. The changes of train are not a problem and will in fact be a relief, if anything.
Flying is quicker and not much more expensive, if at all. But the scenery isn't as good of course.
There is another option - Man in Seat61 won't like it, but it is an option all the same :) - there is a direct bus from Frankfurt to Istanbul. It is a big modern bus with all mod cons such as individual TVs, etc, and runs once a week on Saturdays - cost about €145 http://secure.varan.com.tr/yurtdisi/ - it still takes the best part of two days as it is over 2000 km.
