"Best way" to get to the falls depends on time of year, as well as your budget and preferences. Jul-Aug is peak season; some people prefer to take public transit e.g. www.viarail.ca www.greyhound.ca or www.niagaraairbus.com to avoid parking hassles and traffic frustration. The airbus also goes to BUF.

Car rental certainly gives you more flexilibity. I've noticed that Hertz sometimes has 1way specials: pickup in Toronto, drop in Buffalo.

You can take Maid of the Mist from either Canada or USA side. There's little to do on the USA side of Niagara Falls. Many tourists choose to stay on the Cdn side, where you can view all the falls. Some people do a day trip to the falls from Toronto. Others prefer 1 or 2 nights in the falls to enjoy some of the attractions.

I'd suggest not renting the car until you're leaving Toronto for the Falls. Three reasons: car rentals are cheaper downtown than at the airport, parking for downtown hotels runs $20-$30/night, and you won't need the car for typical sightseeing while you're in Toronto itself.
