Thinking of staying at Renaissance Tuscany il Ciocco Resort in Oct. How far by car to Florence? Rome? What towns are reasonable day trips by car?

It is much too far from Rome and even quite a long way from Florence (well over 2 hours drive and then of course you need to park outside the limited traffic zone and then get into the centre). You could day trip to Lucca. Otherwise you could explore the towns of the Garfagnana such as Barga, Castelnuovo etc. but the roads in this region are slow as it is so mountainous. See the via michelin website for drive times to give you an idea, but bear in mind even these times are optimistic.

Florence is easily done in a day from Ciocco , take the train from Mologno to Lucca connect to Florence , reasonably fast , cheap and no parking/driving in Florence .
