You can probably pick up a cheap and cheerful roadmap at WH Smith at the airport. As for the best route probably the simplest is the one given by theaa.com when you put into it's route-planner boxes, Glasgow Airport to St Andrews. It takes you via Queensferry, but it's not the only option. The AA website is great for route planning and getting an idea of timings, traffic black spots, fuel stops and more:

Arriving from where, as Glasgow is an odd choice if heading for St Andrews. The driving route is basically to drive to virtually the doorstep of Edinburgh Airport then go over the Forth Road Bridge and head towards Dundee until you see signs for St Andrews
And unless it's a weekend you are going to spend a lot of time stationary on the M8 and if jetlagged probably causing the stationary traffic
If you put your start & finish into "google" maps, you will get 3 routes all around the same time (6 minutes difference between the fastest and slowest) - whether or not the times are accurate will depend on the traffic, particularly at the time you will be travelling. It all boils down to your personal choice of what roads you want to drive on.
My personal choice would be via the A91, as it's a more interesting drive with a bit of scenery thrown in (at least on the A91section), rather than the M8 based routes, but you may have other preferences.Edited: 20 October 2010, 19:07

How long will you have been travelling before you reach Glasgow? What is the start point of your flight?
Certainly you are likely to encounter very heavy traffic at that time of the morning heading towards Glasgow and beyond. It does slacken off a little after 9am but it is still busy and you'll need your wits about you from the get-go.
Have you considered any other transport options or are you wedded to the idea of getting a car and heading off to st Andrews the minute you arrive?
We will have a 6 1/2 hour flight arriving at 5am your time, midnight our time.
We had considered other options but the car seemed the better way to go. We will be in Scotland for a little over 3 weeks and plan on doing a bit of sightseeing.
We understand that it is normally a 2 hour drive to St.Andrews so we thought that for the first day we would just get to our rental and relax.
