Travel from Heidelberg to Prague

In September, we will be looking to get from Heidelberg to Prague in a day. Can anyone tell me how best to go about it? I saw a post from several years ago about trains, but the answer was given with letters and numbers that I think would be meaningful to one familiar with the train system. Not to me however. If train is the way to go, I assume there is no direct route. Can one with only English language skills figure out where and how to change as necessary? Any assistance would be greatly appreciated.

Now the easiest option is train to Mannheim and from there by bus.

DB = Deutsche Bahn = German Rail
