we stayed in the hotel boulevard on the kudamm. this hotel is right in the middle of berlin. it's just around the corner of the zoo station with all the trains and buses taking you every where. the hotel is clean with everything you could need and the buffet breakfast was great. you are surrounded by shops and cafes. it really is central. we only paid £50 a night for the room. i have posted a review of this hotel if you need more info.

if you stay here get the number 9 train from the train station {5 minute under cover walk from the airport} leaving from platform 3. get a a,b,c day ticket from info desk at the airport [they speak english] this will cost you 6.50 euros and you can travel anywhere in a,b,c, zone in berlin untill midnight that day. do not get a taxi as this will cost you over 30 euros each way.

get the train to zoologicher station, the hotel is one block away.

minor correction: day tickets are valid till 3am next day

However with 3 nights the 72 hours Berlin Welcome Card may be the even better option

Zoo station area is close to one of the main shopping areas (Kurfürstendamm), but there are not that many sights in this area. You may also consider the Mitte district. But generally you will need public transport to get around. Berlin is huge.

A good idea is to start with one of the offered walking tours:

Note: also for most walking tours you will need a public transport ticket

Most berlin visitors either stay in Mitte (where a majority of sights are) or in Charlottenburg (Ku'Damm area where there is one of Berlins major shopping street but less sights). Berlin is quite widespread therefore you will have to use public transport anyway. Therefore it doesn't matter so much where you stay as long as it is within Central Berlin and has an underground or S-Bahnstation near by. Hotels in Mitte are sometimes a bit more expensive than in other Central areas. I can't really recommend a hotel itself so.

As someone living in Berlin longterm I don't like Mitte so much. It is quite touristy and sometimes I find it hard to find nice bars/restaurant there which are not too overpriced for what is offered. Anyway, I can see what many tourists like in Mitte.
