Yes, there are trains from Rome to Fondi-Sperlonga - Just enter Rome and Sperlonga on the Trenitalia website – The trip would take 01h10.

I would also follow that link to piazzoli.it, click on Orari, and click the arrow next to Orario di Sperlonga. (The .pdf is 26 Mb, despite being a single page, so I'd avoid downloading it!)
The bus schedule varies depending on time of year and is fairly inconvenient. It's best to look at the schedule and organize your arrival and departure accordingly. The buses don't pass often, and if you miss it there's only one taxi. (It's not even a legitimate taxi, despite his claims, but he's reliable. Look for an Egyptian guy with an SUV - he asks €30-35 but has been known to take 6 people for €25.) The same goes for the ride back: plan ahead!

In the summer time the bus will be absolutely packed, so forget the concept of personal space. I've never heard of anyone being robbed there, but in a tight space it's always smart to watch your wallet.
Also, people will not be afraid to push and cut in front of you while waiting for the bus (adults and otherwise). I've heard a fair share of swearing during the process. Don't let it unsettle you. It's all part of the "fun" of fighting your way to the beach in the summer. (It's worth it, trust me!)
Also, watch for what's written on the bus as one goes to Fondi and the other to Sperlonga. Make sure you get the right one!
You can buy tickets for the bus in the bar at the station or on the bus itself.
Edited: 9 years agoIf you'd like to avoid the bus, your hotel could always arrange a transfer for you.

It just depends on the time of year and a bit of luck. I go there often, and in the crazier times I usually find tourists who aren't quite sure what's supposed to happen. It's better to be prepared, and any hassle is worth it!
