We went from termini station to Vatican by Metro, fastest way that I know of.

We stayed a few blocks from Termini station, and took the metro (not from Termini itself, but from a stop between our apartment and Termini). We purchased tickets from a newstand just by the entrance to the metro. Very easy.

If you're going to the museums the fastest way is probably the metro. If going to the basilica you should probably catch the bus. Your hotel can tell you which one to take.

The Metro (line A) is much quicker than the bus; it's the best way to get from Termini to the Vatican.

It may be quicker, but I like the bus better.
