No one can give you an order or suggest a route before you tell us what you want to see. You haven't listed the sites you're interested in. Once you list them we can help.

First, you'll need a good map.

From the termini head to Santa Maria Maggiore, the colosseum, the circus maximus, piazza Venezia, trevi fountain, Piazza Barberini and then take the metro to the Vatican museums. I doubt you'll have time for the museums but you should have time to look around the basilica.

The route is pretty easy to walk with the exception of getting to the Vatican which is why you should take the metro there.

I agree, as you won't be able to do even all the top-rated sights/activities listed on tripadvisor.com/Attractions-g187791-Activiti… so you'll need to decide what YOUR priorities are for the time you have. People could waste time telling you how to get to their favourite place only to find that it isn't the sort of thing you want to do at all!

I suggest you take a look at the examples on http://www.rome.info/itineraries/ - reidsitaly.com/destinations/lazio/rome/sight… - and Go Lists for Rome on TA: tripadvisor.com/GoListOverview-g187791-Rome_… - after that, you could ask more specific questions on how to get from A to B.

For example, you could start by walking (about 30 mins) from Termini to the Colosseum, which is close to the Forum, Palatine and Capitoline hills/museums. Then maybe the Pantheon (20 mins walk from the Capitoline), and Piazza Navona, then the Spanish Steps and Trevi Fountain. But if you want to visit the Vatican museums, you might want to take the metro straight there. It all depends on what YOU want to do!
