There are parts of Puglia very well served by train and bus...it depend what town or area you have in mind?

But yes - it is possible. You'll be slightly more limited as to where you can get to easily, and sometimes the train and bus timetables can be quite frustrating, but you can get around alright, it's just that the times you travel will be dictated by some slightly idiosyncratic timetables :) It depends on exactly where you're going. The small local trains (eg between Lecce and Gallipoli / Otranto) don't appear on the Trenitalia website, as they're run by a different company, Ferrovie del Sud Est - their website is www.fseonline.it (only in Italian, unfortunately), and that will give you local train AND local bus times. Further north there are a couple of other local train companies - Ferrovie del Nord Barese, which you probably won't need to use, and the Ferrovie Apulo Lucane (http://www.fal-srl.it/), which connects Matera and Bari, among other places.
