The R train won't go to Barclays Center. You'll have to change trains. You can take the R one stop from Queens Plaza to Lexington Avenue and, on the same platform, get the N or Q to Barclays Center.

I think your hotel is actually closer to 33rd Street on the 7 train. If you want to leave from there, then take the 7 to Grand Central and change for the downtown 4 or 5 train to Barclays Center.

The R train DOES go to Barclay's Center, but it no longer runs from Manhattan to Brooklyn, because the tunnel was severely damaged in the hurricane last year. It runs in two separate sections, one section from lower Manhattan to Queens, and one section from downtown Brooklyn to Bay Ridge. So taking teh R train (or 7 train) from your hotel to another subway line makes the most sense.

On weekends, the R train does run straight through, but across the bridge, instead of through the tunnel. Another line will still be faster, because the R is a local train in Manhattan and Brooklyn.
