Hi my wife and I are trying to figure out the best route from Lanquin to Placencia. Any advice?
Since the trip will take days, it largely comes down to what you want to see and where you want to stay enroute. For example, you could go:
:: from Lanquin to Rio Dulce on local buses and shuttles, east to the Caribbean, and then via boat to southern Belize
:: back to Guatemala City to head by bus or flight in either direction

No direct transport between the two, but a variety of indirect options. A couple of suggestions below.
If you are up for some scenic adventure you could take local micro bus transport on the back roads to El Estor and on to Fronteras (Rio Dulce). Spend a few days on the Rio and then water taxi to Punta Gorda, Belize. Then bus or fly to Placencia.
If you just want to get there, then one option is bus it back to Coban, then bus to Puerto Barrios, water taxi to Punta Gorda.
Whichever option you chose, short of a private charter flight, you are almost certainly going to need to stay overnight somewhere on the route, but why hurry?
I take it you would like to visit Semuc Champey and then head to Belize. Would you like to see Tikal? Because it would make a nice stopover on a nice road trip to Belize on good roads. Rio Dulce and Livingston are great but unless you plan to spend some time there specifically, that route to Placencia is long, arduous and, frankly, uncomfortable on various busses and boats on often very rough waters.
As the other contributors noted, there is no quick way from Languin to Placencia.
If it's simply a quick snorkelling trip you are after, you may be better off simply heading to Rio Dulce and getting a package to the Cayes.
Otherwise, Lake Peten Itza is glorious (didn't make it to Tikal, going back in March) and it's a great stop on the way to Belize.
