It is a very pleasant drive but you can also consider taking the train from Izmir to both Selçuk and Denizli. http://www.tcdd.gov.tr/tcdding/index.htm

It depends what you mean by "Izmir". Personally I find that a very difficult city to navigate, but once you're out onto the motorway it's pleasant and fast. If you mean the airport, then there's no problem at all because it's well south of the city.

When you join the motorway be sure to follow signs for "AYDIN". Then Ephesus is indicated by the usual brown sign for ancient sites.

Driving near Selcuk / Ephesus is fun. The road onward to Aydin and Denizli is fine - dual-carriageway and flat after the mountain South of Ephesus. Too many traffic lights for my liking.

As Borus mentions, the train is now a very good option.

Renting a car and driving between those 3 places is prety doable for an accomplished driver.

The only danger would be yourself. If you are in control of the vehicle and follow the road you won't encounter any dramas.
