You can fly from Lima to Cusco. Takes about 90 minutes.

Then bus or train to Puno (at L Titicaca).

The nearest airport to Puno is at Juliaca. But the journey between Puno and Cusco is quite interesting, so it may be worth returning overland to Cusco, which is an interesting city and has frequent flights to Lima.

Both Cusco and Puno are high and the altitude may affect you. It may be worth acclimatising somewhere lower for a couple of days, eg the Sacred Valley / Machu Picchu.
