Have a look here:

enjoy-patagonia.org/ushuaia-how-to-get-there…

"As there are no buses operating from El Calafate to Ushuaia, you can either go to Río Gallegos by bus and take a plane to Ushuaia; or go to Puerto Natales and then to Punta Arenas (Chile) by bus, where you should take another bus to Ushuaia. Interlagos y Quebek Tours provide buses from El Calafate to Río Gallegos every day all the year round. The trip takes 4 hours. The companies Cootra R.T. Ltda. and Bus Sur connect El Calafate with Puerto Natales (Chile) in a regular way. From Puerto Natales to Punta Arenas there are buses every hour from 8 a.m. to 8 p.m. all the year round. The trip takes 4 hours."

It is quicker to go by plane.
