I would stay in Asheville and use that as your base. You might like ALoft or Hotel Indigo. They are both conveniently located downtown, easy to walk to dinner, etc.
Based on where you're visiting while you're in Asheville, somewhere in the middle, like downtown Asheville or close to downtown would be a good idea. Take a look at www.exploreasheville.com for some ideas.
For a slightly different option and higher in elevation, so cooler. Check out the off the beaten path, northeast of the Asheville, 2 miles off of the Blue Ridge Parkway - Sourwood Inn (www.sourwoodinn.com) 45 minutes from Mt. Mitchell, head down the BRPKWY to 74A out to Lake Lure, and about 15 miles to the Biltmore Estate (but will take about 30 minutes due to windy mountain road that the inn sits on (5.5 miles). we had very good time/service at Homewood Suites in Asheville and had good access to all the areas you mentioned. The suites with two queen beds were large and comfortable
