The Greyhound bus station is on Stewart. It is only a few blocks north of the Paramount Theater and the Convention Center, which are on the east end of Pike/Pine in downtown. So it is not hard to get to. If you're there during office hours, go across the street to Dilettante to get a custom hot chocolate...

I don't know where the Bolt Bus picks up.

BoltBus is going to be the cheapest. Tickets there start at $1, but good luck getting that price. If they sell out for a particular time slot, then they start selling regular greyhound tickets. They pick up from 5th Ave S & S King St Seattle, WA 98104 and arrive at 1150 Station Street Vancouver, BC V6A4C7

As suggested Boltbus is probably cheaper, and Amtrak is more comfortable.
