It's a beautiful, beautiful place to be at. Well worth the trouble, in my opinion.
I took the bus from Rethymnon -> Preveli. It's the same bus that goes to Plakias but stops over at Preveli. You can check KTEL's website for the route and price. Do take note that the bus stop is at an obscure location and you MUST inform the bus driver that you're getting off at Preveli. The bus stop you're looking for is immediately after the Preveli Monastery stop.
There's no indication that it's a bus stop - But the bus will stop at the mouth of the road heading towards a carpark that leads to Preveli Beach.
Once at the "bus stop", it's a 30 - 45 minute walk to the beach. You'll have to walk along the road, past the car park to a cliff and down few hundred steps in order to get to Preveli. View from above is magnificent, so don't forget to snap them photos!
Renting a car and driving there would infinitely be easier - but you've still got to climb up and down said few hundred steps.
Well worth the trip but only if you're up for the "hike".
If you're taking the bus back to Rethymnon, don't forget to bring with you the bus schedule and be there waiting for the bus, else you get left stranded at the "bus stop". Give the bus +/- 15 minutes from said schedule to arrive and it will. (Don't freak out the way I did)
The alternative route would be to take a ferry to Plakias?
